CJC-1295

CJC-1295 is a synthetic analog of growth hormone-releasing hormone (GHRH) with 30 amino acids. It is available in two forms: with and without Drug Affinity Complex (DAC). It has been studied in animal models for its effects on pulsatile growth hormone release.

Research Summary

CJC-1295 is a modified GHRH analog designed for increased stability and extended half-life compared to native GHRH. The version with DAC (Drug Affinity Complex) utilizes a maleimido derivative of lysine that binds to circulating albumin, further extending its biological activity. Research in animal models has demonstrated significant and sustained elevations in growth hormone and IGF-1 levels. The non-DAC version (also known as Modified GRF 1-29) maintains the pulsatile release pattern more closely resembling physiological GH secretion.

Mechanism of Action

CJC-1295 binds to GHRH receptors on anterior pituitary somatotroph cells, activating cAMP/PKA signaling cascades that stimulate growth hormone synthesis and release. The DAC modification allows albumin binding, extending plasma half-life from minutes to days. It preserves the pulsatile pattern of GH release when used in conjunction with GH secretagogues.

Published Animal Studies

  • [1]Studies in animal models demonstrated sustained elevation of GH levels for 6-8 days following single administration of DAC-modified CJC-1295 (Jetté et al., 2005)
  • [2]Rodent studies showed increased IGF-1 levels and enhanced growth parameters with chronic administration
  • [3]Primate research demonstrated dose-dependent GH release with preservation of pulsatile secretion pattern
  • [4]Combination studies with GHRP-type peptides in animal models showed synergistic effects on GH release (Bowers et al., 1990)

In-Vitro Research

  • Pituitary cell cultures demonstrated receptor binding affinity and cAMP activation comparable to native GHRH
  • Stability studies showed dramatically increased resistance to DPP-IV enzymatic degradation
  • Albumin binding assays confirmed high-affinity association of DAC-modified CJC-1295 with serum albumin
